Reading a retatrutide COA: what to check on a triple-agonist peptide
Retatrutide threads mostly cover what the molecule does. Less discussed is how
to read the paperwork that comes with a vial, and retatrutide has a couple of
features that make its certificate worth a closer look than a short peptide’s.
It’s a 39-residue synthetic peptide carrying a fatty-acid modification. Two
consequences on the analytical side:
1. Length drives the impurity profile. Longer synthetic sequences accumulate
more deletion and truncation products during synthesis than something short
like BPC-157 does. Those show up on a chromatogram as smaller peaks eluting
near the main one. A summary line reading “99.2%” tells you nothing about
whether they’re there — ask for the actual trace, not just the number.
2. The acylation has to show up in the mass. Mass spectrometry confirms
identity by matching observed molecular mass against theoretical mass for the
sequence. The lipid modification is part of that theoretical figure. An
observed mass that matches the unmodified backbone instead is a different
compound than the label says, and it’s the single most useful check on the
certificate.
Beyond that, the standard four:
– Batch number on the certificate matches the vial in your hand. A COA reused
across every subsequent lot describes material you don’t have.
– HPLC method named — column, gradient, detection wavelength. A purity claim
with no method behind it is a number without a procedure.
– Testing lab named and independent. In-house results are a supplier grading
their own work.
– Test date after the production date.
On storage: lyophilized is the stable form, kept cold, dry and dark. Once
reconstituted the usable window shortens considerably — refrigerate, protect
from light, and avoid repeated freeze-thaw cycles, which are harder on peptide
integrity than cold storage itself.
